Led Xenon
A UV Visible Spectrophotometer is the instrument which is used to measure the intensity of light before and after passing through a sample – the ratio of one to the other is called transmittance and is expressed as a percentage. From transmittance, absorbance can be calculated.
While the most common spectrophotometers are used in the UV and visible regions of the spectrum, there are also instruments which operate in the near-infrared region.
The basic parts of a spectrophotometer are a light source, a sample holder, diffraction grating or monochromator (used to separate light by wavelengths) and a detector. The radiation source is typically a deuterium lamp, which provides continuous light over the ultraviolet region. More recently, light emitting diodes (LED) and Xenon lamps have come into use for visible wavelengths. The detector is normally either a photodiode or a CCD. Photodiodes are used with a UV visible spectrophotometer which uses a monochromator, while diffraction gratings are used with the CCDs, which collects light of different wavelengths on different pixels.
UV Visible spectrophotometers are routinely used in the quantitative determination of solutions of transition metal ions and highly conjugated organic compounds. Since electrons within the metal atoms can be excited from one electronic state to another, these solutions can be colored; the color is affected by the presence of certain anions or ligands. The color of a dilute solution of copper sulfate, for example is a light blue. Adding ammonia intensifies the color and changes the wavelength of maximum absorption of the solution.
Organic compounds also absorb light in the UV and visible regions of the electromagnetic spectrum. The solvents for these determinations with a UV visible spectrophotometer are water for water soluble compounds or ethanol for organic-soluble compounds. Organic solvents may have significant UV absorption, but this does not mean that these solvents are universally suitable for use in every application involving UV visible spectrophotometers, especially since ethanol absorbs very poorly at most wavelengths.
Solvent polarity and pH can also have an effect on the absorption spectrum of organic compounds. The Beer-Lambert law states that the absorbance of a solution is directly proportional to the concentration of the absorbing species in the solution and the path length. Consequently, for a fixed path length, a UV visible spectrophotometer can be used to determine the concentration of the absorber in a solution. It is necessary to know how quickly the absorbance changes with concentration. This can be ascertained from reference tables of molar extinction coefficients or from a calibration curve.
UV visible spectrophotometers may be used as a detector for high performance liquid chromatography (HPLC). The presence of an analyte gives a response which can be assumed to be proportional to the concentration. Visible region spectrophotometry is used extensively in colorimeter science. Ink manufacturers, printing companies, textiles suppliers and many other companies need the data provided by a UV visible spectrophotometer. This data can be used to test a new batch of colorant to determine whether or not it meets specifications such as ISO printing standards.
Traditional UV Visible Spectrophotometers cannot detect if a colorant or the base material has fluorescence. In respect to inks, this can make it difficult to manage color issues if any of the printing inks happen to be fluorescent – if a given colorant is fluorescent, a bi-spectral fluorescent spectrophotometer can be used instead.
